40 µl is required for each test. However, the total sample volume needed is dependent on the type of sample tube used and dead volume for that specific tube.

If the Phadia 250 instrument dilution is used the sampling amount depends on the dilution factor:
- 1:10 (one-step dilution) = 9 µ l of neat sample
- 1:50 or 1:100 (two-step dilution) = 20 µ l of neat sample
